CELL CULTURE QUALITY CONTROL: Improving cell-based assays

There are many different factors which can adversely affect cell culture and cell-based assays including; a.) human error, subjectivity and inconsistency b.) environment c.) reagent and labware variability and d.) the time-dependent nature of biology itself. IncuCyte provides a simple, non-invasive, time-lapse method of archiving and quantifying cell growth inside your cell culture incubator. This capability can be used to standardize cell culture procedures, document growth and morphology over time, isolate variables and optimize cell-based assay performance.

NEW CELLPLAYERTM ASSAYS: Kinetic assays in your lab or ours

Biological processes are, of course, time-dependent. We believe that providing assay read-outs which provide more than a single snapshot in time is essential to understanding the process itself. Utilizing our IncuCyte technology and our biotechnology team, we have developed live-cell imaging assays designed to provide a kinetic read-out to what previously have been single read “end-point” assays. Our CellPlayerTM Assays allow you to:

  • Resolve transient signaling events
  • Determine the time course of drug action
  • Provide insight into drug mechanism
  • Optimize assay timing (data sampling, compound addition)
  • Increase the signal to background ratio

 

NEW CELLPLAYERTM ANGIOGENESIS ASSAY

Angiogenesis, the formation of new blood vessels from pre-existing vasculature, is a key biological process involved in numerous physiological and pathological events including wound healing, tumor growth and diabetic retinopathy. We have developed a kinetic, primary cell, co-culture assay model by labeling endothelial cells with GFP, reading with IncuCyte FLR, and integrating custom analysis metrics which quantify tube formation and branching over time. The kinetic readout enhances mechanistic pathway analysis, assay dynamic range and optimizing compound addition.

NEW CELLPLAYERTM MIGRATION ASSAY

Cell migration is a fundamental, yet complex biological process involved in almost every aspect of human biology.  In a healthy individual, it is critical to our immune response and wound healing.  Cell migration also plays critical roles in disease, specifically in the areas of asthma, rheumatoid arthritis, tumor growth, as well as atherosclerosis and vascular disease.  We have developed an automated, 96-well, kinetic, non-labeled, cell migration assay around our IncuCyte platform.   Using this assay we can quantify cell migration in time, without labels, and without removing cells from the incubator.

KINETIC CELL PROLIFERATION

IncuCyte can be used to follow kinetic cell proliferation in a number of ways. Using our integrated confluence algorithm as a surrogate for cell number, researchers can track the effects of drugs on cell proliferation without labeling. When absolute cell count is important, live-cell dyes such as Vybrant® DyeCycle™ Green or GFP labels in conjunction with integrated segmentation and object counting can provide a direct readout of cell number vs. time.
